Design: Eukaryotic mRNA was enriched by magnetic beads with Oligo (dT) by binding A-T to the ployA tail of the mRNA. Subsequently, break the mRNA into short fragments. The mRNA was used as a template to synthesize a strand of cDNA with six random bases and then the buffer, dNTPs and DNA polymerase I were used to synthesize the double-stranded cDNA. AMPure XP beads were then added to purify double-stranded cDNA. PCR-enrichment was performed to obtain the final cDNA library.

This work presents a combination of experiment and analysis aimed to describe the priority tolerant ability of Z. rouxii which caused food spoilage. These knowledge will provide clues or methods to decrease the food spoilage caused by sugar and salt resistant-yeast species.
